Dehydroepiandrosterone (DHEA) is a multi-functional neurosteroid that has been implicated in a variety of biological effects in humans and other mammals. The mechanism of action of DHEA has not been fully elucidated, although it is under investigation as a replacement therapy for humans with DHEA Insufficiency based on reports suggesting beneficial effects ranging from anti-depression to neuroprotection. Controversy remains about the relative contribution of DHEA and its metabolites in its biological effects. This dissertation focuses on a comparative study of the genomic effects of DHEA and its metabolites in vitro, and identification of genes regulated by DHEA in vivo. The results indicated that DHEA, androstenediol and androstenedione produced typical genomic effects, including promoting AR-GFP intracellular trafficking and formation of nuclear clusters in COS-7 cells, and conferred AR transcriptional activity in a dose-dependent manner in transiently transfected CV-1 cells. In contrast, DHEAS, 7alpha-OH-DHEA, 7beta-OH-DHEA, and 7-oxo-DHEA were essentially without androgenic activity. The results confirmed that DHEA is intrinsically androgenic and demonstrated significant differences in androgenic activity among DHEA and its metabolites. In the hypothalamus, DHEA, like the strongest androgen DHT, significantly up-regulated the expression of two important neuropeptides: hypocretin (Orexin) and pro-melanin concentrating hormone precursor (Prepro-MCH), and delta type protein kinase C (PKCdelta), and down-regulated the expression of deleted in bladder cancer chromosome region candidate 1 (DBCCR1) and chitinase 3-like 3 (Chi3l3). Interestingly, DHT down-regulated the expression of PKCS in the hippocampus. The differential expression of Prepro-MCH, orexin, and PKCdelta was further confirmed by real-time RT-PCR. MCH and orexin are involved in the regulation of appetite and energy homeostasis, and PKCdelta and DBCC1 are involved in regulation of cell growth, proliferation or survival. More importantly, the functions of these genes may explain the multiple effects of DHEA in humans and rodents. The findings may be of clinical significance and provide a new level of detail concerning the mechanism of action of DHEA in the central nervous system.
