The search for new Pt(II) metal drugs is underpinned by an in-depth understanding of the factors controlling the bio-reactivity of the Pt(II) pharmaceuticals. An emerging class of compounds which has shown improved antitumour activity is the multinuclear Pt(II) complexes, epitomized by the favourable reactivity profile of BBR3464 from Prof. Farrell's labs. Kinetic data of multinuclear Pt(II) compounds is not extensive. This book, reports the substitution reactions of bis(2-pyridylmethyl)amine-chelated dinuclear Pt(II) complexes linked by diamine bridges. The reactions were studied under pseudo first-order conditions as a function of concentration of nucleophiles and temperature using stopped-flow and UV-visible spectroscopic techniques. In general, the substitution of the coligand by the sulfur containing nucleophiles proceeds via a two-step reaction pathway which is associatively activated. In the studied sets of Pt(II) dinuclears, substitution of the coligands is controlled mainly by steric effects, derived from the molecular conformations of the metal complexes and to a lesser extent by electronic effects. Experimental data is well supported by Quantum mechanical calculations.

B-cell chronic lymphocytic leukemia (CLL) is considered a single disease with extremely variable course, and survival rates ranging from months to decades. It is clear that clinical heterogeneity reflects biologic diversity with at least two major subtypes in terms of cellular proliferation, clinical aggressiveness and prognosis. As CLL progresses, abnormal hematopoiesis results in pancitopenia and decreased immunoglobulin production, followed by nonspecific symptoms such as fatigue or malaise. A cure is usually not possible, and delayed treatment (until symptoms develop) is aimed at lengthening life and decreasing symptoms. Researchers are playing a lead role in investigating CLL's cause and the role of genetics in the pathogenesis of this disorder. Research programs are dedicated towards understanding the basic mechanisms underlying CLL with the hope of improving treatment options.

For the past 40 years, metal-based drugs have been widely used for the treatment of cancer. Cisplatin and follow-up drugs carboplatin (ParaplatinTM) and oxaliplatin (EloxatinTM) have been the gold standard for metallodrugs in clinical settings as antineoplastic agents. While effective, these drugs (either alone or in combination therapy) have faced a number of clinical challenges resulting from their limited spectrum of activity, high toxicity leading to significant side effects, resistance, poor water solubility, low bioavailability and short circulating time. In the past 10 years, various unconventional non-platinum metal-based agents have emerged as a potential alternative for cancer treatment. These compounds are highly effective and selective in cancers resistant to cisplatin and other chemotherapeutic agents. Research in this area has recently exploded with a relevant number of patents and clinical trials, in addition to reports in scientific journals. Furthermore, in parallel to the synthesis of coordination and organometallic compounds comprising many different metals and unconventional platinum-based derivatives, researchers are focused on optimizing mechanistic and pharmacological features of promising drug candidates. This Special Issue aims to highlight the latest advances in anticancer metallodrugs with a focus on unconventional anticancer agents, as well as novel activation, targeting and delivery strategies aimed at improving their pharmacological profile.
