Die Studie mit dem Titel „Investigation of bio-organic systems by means of optical techniques“ befasst sich im technischen Teil mit dem Aufbau und der Inbetriebnahme eines neuartigen Ramanspektrometers. Es wurden die wesentlichen Leistungsparameter des Systems untersucht und mit Erwartungen aus Simulationen verglichen. Ausserdem wurde eine Systemintegration mit Lichtquellen, Strahlengang und Justageprozeduren durchgeführt. Weiterhin beschäftigt sich diese Arbeit mit den Möglichkeiten optischer Untersuchungsmethoden bezüglich keratinbasierter bio-organischer Materialien. Mittels spektroskopischer Ellipsometrie wurde neben allgemeinen Charakterisierungsuntersuchungen eine Studie der Hydrationsdynamik von Fingernägeln durchgeführt. Die Ergebnisse dieser Studie wurden mit einem Modell bestehend aus ,,freiem`` und ,,gebundenem`` Wasser erklärt. Die Ellipsometrie wurde auch für eine in-vivo Tapestripping Studie verwendet, welche ein Tiefenprofil des stratum corneums lieferte und aufzeigte, das Depolarisationseffekte eine grosse Rolle bei der Ergebnisinterpretierung spielen und in Zukunft weiter untersucht werden sollten. Das neue Ramanspektrometer wurde ebenfalls für biophysikalische Untersuchungen eingesetzt. Im tiefen UV bei 244 nm wurde eine Zerstörungsstudie an Einzelhaaren und eine Studie an Hautbiopsien durchgeführt. Motivation für Messungen in diesem Wellenlängenbereich sind Fluoreszenzuntersuchungen im Sichtbaren sowie eine ebenfalls durchgeführte Raman-Resonanzstudie an destilliertem Wasser im Wellenlängenbereich zwischen 631 und 237 nm. Während der Studien wurden die technischen Grenzen beider optischer Aufbauten offenbar und führten u. a. zu dem Design eines neuen Periskops für das Ramanspektrometer, um die Probenpositionierung zu verbessern. Außerdem wurden durch die Ellipsometriemessungen Grundlagen zum Aufbau eines neuartigen, speziell an biologische Systeme angepassten Ellipsometers gelegt.
