The central theme of this book is that systems of cell-cell signalling via nerves, hormones, local mediators and growth factors are not distinct phenomena, but branches of one general mechanism. These topics therefore can and should be discussed in an integrated manner, and the division of cell signalling studies into separate pigeonholes such as neuroscience, endocrinology or cancer biology is unnecessary, if not counterproductive. I also believe it to be unfortunate that there is not a collective term to describe neurotransmitters, hormones, local mediators and growth factors, other than clumsy phrases such as "extracellular signal molecule". The lack of a short and distinctive word for these entities genuinely hampers people from thinking about them in an integrated way. Having decided that it was presumptuous to invent a new term, I have chosen in this book the term first messenger to cover all types of extracellular signal molecule, because of the widespread acceptance of the term second messenger to represent the intracellular signal molecules that are produced in response to many of them. I have given the book the title "biochemical messengers", which is a global term to cover both first and second messengers. The impetus for writing the book came, as must often be the case, when I had to put together a course on cell-cell signalling for biochemistry students at the University of Dundee.

An essential element in the development and functional integrity of all organisms is intercellular communication. This is achieved by the secretion of soluble messenger molecules which subsequently interact with receptor-effector pathways in the responsive cells. Hormones are traditionally defined as chemical messengers synthesized by endocrine glands. Unlike hormones produced by endocrine glands, growth factors are hormone-related substances produced by many tissues and play an important role in controlling growth and development. While the exact physiological roles of growth factors have yet to be elucidated, they play important roles in the regulation of cellular proliferation and/or differentiation during ontogenesis, growth and differentiation. During recent years there has been a substantial increase in research related to peptide growth factors, their receptors, and modes of action. With the discovery and characterization of numerous growth factors, it became clear that these growth factors had multiple features in common with classic hormones as well as with oncogenes. Furthermore, there are distinct families of growth factors based either on structural or functional similarities.

Endocrinology is a field in which enormous advances have been made in the last decade; the rate of discovery of new hormones, hormone-like molecules, receptors, and mechanisms of action is continually advancing. The development of techniques in immunology and molecular biology has led to the possibility of describing in detail the gene structure of many of the compounds involved in hormonal systems. Remarkable homology has been shown between oncogene products and various components of the endocrine network, leading to the asser tion that deregulation of hormonal function is involved in the generation and/or development of cancer. We now know that the central nervous system is both a target and a production site of many hormonal products, and that hormones, neurotransmitters, growth factors and immunopeptides all act through similar mechanisms. The only second messenger known ten years ago was cAMP; today calcium, derivatives of membrane phospholipids, and protein kinases are also known to be mediators of hormone action. The very concept of hormonal systems has been expanded to include not only endocrine secretions but also para- and autohormones and their mechanisms of action; an understanding of their functions will be central to the immediate future of medicine. The discovery of hormonal molecules and endocrine interactions and the subsequent understanding of hormone related pathophysiology has led to the development of new strategies in medical treatment such as fertility control and the management of diabetes.
